Âñòóïëåíèå. Èçó÷åíèå ìåõàíèçìîâ íåéðîãóìîðàëüíîé ðåãóëÿöèè ôóíêöèîíàëüíîãî ñîñòîÿíèÿ íåðâíî-ìûøå÷íîé ñèñòåìû ïî-ïðåæíåìó îñòàåòñÿ ïðåäìåòîì ìíîãî÷èñëåííûõ ýêñïåðèìåíòàëüíûõ èññëåäîâàíèé [3, 4, 6, 9, 12, 15]. Îñîáîå ìåñòî â äàííîé ïðîáëåìå îòâîäèòñÿ ùèòîâèäíîé æåëåçå.  ðàáîòàõ ðÿäà àâòîðîâ ïîêàçàíî [3, 5, 7, 9, 14], ÷òî ïðè íàðóøåíèè òèðåîèäíîãî ñòàòóñà íàñòóïàþò ìíîãî÷èñëåííûå èçìåíåíèÿ ñî ñòîðîíû ðàçëè÷íûõ çâåíüåâ íåðâíîìûøå÷íîãî àïïàðàòà, ÷òî ìîæåò ñëóæèòü îñíîâîé êàê óëó÷øåíèÿ, òàê è óõóäøåíèÿ ôóíêöèîíàëüíûõ ïàðàìåòðîâ ñêåëåòíîé ìûøöû [1, 8]. Îäíàêî îñòàåòñÿ íåâûÿñíåííûì âîïðîñ î õàðàêòåðå âëèÿíèÿ ýêñïåðèìåíòàëüíîãî ãèïåð- è òèðåîòîêñèêîçà íà ïàðàìåòðû, îòðàæàþùèå ìîáèëüíûå õàðàêòåðèñòèêè ñîêðàòèòåëüíîãî àêòà, â ÷àñòíîñòè åãî ïåðâûå ñòàäèè. Ðàáîòàìè ìíîãèõ àâòîðîâ ìåòîäàìè ôèçèêîõèìè÷åñêîãî àíàëèçà ïîêàçàíî, ÷òî ïðè ñîñòîÿíèè ãèïåðòèðåîçà èçìåíÿþòñÿ ïëîòíîñòü è ìîáèëüíûå õàðàêòåðèñòèêè Nà+-êàíàëîâ [13], àêòèâíîñòü Nà+/ Ê+-ÀÒÔàçû ïëàçìàòè÷åñêîé ìåìáðàíû è ìèîçèíîâîé ÀÒÔàçû ñîêðàòèòåëüíîãî àïïàðàòà [13], ñòåïåíü ñðîäñòâà àêòèíîâûõ íèòåé ê èîíàì Ca2+, ïëîòíîñòü è ôóíêöèîíàëüíîå ñîñòîÿíèå Ca2+-êàíàëîâ ìåìáðàíû ñàðêîïëàçìàòè÷åñêîãî ðåòèêóëþìà [10], àêòèâíîñòü åãî Ca2+- ïîìïû [11, 14] è äð. Òàêèì îáðàçîì, òèðåîòîêñèêîç âûçûâàåò ìíîãî÷èñëåííûå èçìåíåíèÿ ñî ñòîðîíû ìíîãèõ çâåíüåâ íåðâíî-ìûøå÷íîãî àïïàðàòà, ÷òî ÿâëÿåòñÿ ïðîÿâëåíèåì èõ ïàòîôèçèîëîãè÷åñêîãî ýôôåêòà. Îäíàêî, íåñìîòðÿ íà äîñòàòî÷íî ïîëíóþ õàðàêòåðèñòèêó ýôôåêòà òèðåîèäíûõ ãîðìîíîâ íà ðàçíûå ñòîðîíû ôóíêöèîíàëüíîãî ñîñòîÿíèÿ ñêåëåòíîé ìûøöû, ìíîãèå àñïåêòû ïðîáëåìû îñòàþòñÿ íåäîñòàòî÷íî èññëåäîâàííûìè.  ïåðâóþ î÷åðåäü ýòî êàñàåòñÿ õàðàêòåðà çàâèñèìîñòè ìåæäó ñòåïåíüþ íàðóøåíèÿ òèðåîèäíîãî ñòàòóñà è âûðàæåííîñòüþ ïàòîôèçèîëîãè÷åñêèõ ìåõàíèçìîâ, ðàçâèâàþùèõñÿ â íåðâíî-ìûøå÷íîé ñèñòåìå. Öåëüþ ðàáîòû ÿâèëîñü âûÿñíåíèå â óñëîâèÿõ in situ õàðàêòåðà ñâÿçè ìåæäó ïîêàçàòåëÿìè, îòðàæàþùèìè ìîáèëüíûå ïàðàìåòðû ñîêðàùåíèÿ ïåðåäíåé áîëüøåáåðöîâîé ìûøöû áåëûõ êðûñ ñ ðàçëè÷íûì òèðåîèäíûì ñòàòóñîì, ñ îäíîé ñòîðîíû, è óðîâíåì öèðêóëèðóþùåãî ñâîáîäíîãî òðèéîäòèðîíèíà – ñ äðóãîé. Îáúåêò è ìåòîäû èññëåäîâàíèÿ. Ýêñïåðèìåíòû âûïîëíåíû íà 80 âçðîñëûõ áåëûõ êðûñàõñàìöàõ ìàññîé 282 ± 1,1 ã. Ñîäåðæàíèå æèâîòíûõ è ýêñïåðèìåíòû ïðîâîäèëèñü ñîãëàñíî ïîëîæåíèé «Åâðîïåéñêîé êîíâåíöèè î çàùèòå ïîçâîíî÷íûõ æèâîòíûõ, êîòîðûå èñïîëüçóþòñÿ äëÿ ýêñïåðèìåíòîâ è äðóãèõ íàó÷íûõ öåëåé» (Ñòðàññáóðã, 1985), «Çàãàëüíèõ åòè÷íèõ ïðèíöèï³â åêñïåðèìåíò³â íà òâàðèíàõ», óòâåðæäåííûõ Ïåðâûì íàöèîíàëüíûì êîíãðåññîì ïî áèîýòèêå (Êèåâ, 2001). Âñå æèâîòíûå áûëè ðàçäåëåíû íà äâå ãðóïïû. Ïåðâîé ãðóïïå êðûñ (30 æèâîòíûõ) åæåäíåâíî â òå÷åíèå 4-õ äíåé ïîäêîæíî ââîäèëñÿ òðèéîäòèðîíèí â äîçå 15 ìêã/êã (ãèïåðòèðåîèäíàÿ ãðóïïà). Äëÿ îïûòà æèâîòíûå èñïîëüçîâàëèñü íà ñëåäóþùèé äåíü ïîñëå îêîí÷àíèÿ èíúåêöèé ãîðìîíà (5-å ñóòêè). Âòîðàÿ ãðóïïà êðûñ (50) áûëà êîíòðîëüíîé. Ðåçóëüòàòû ïîêàçàëè, ÷òî âåëè÷èíà ïîòðåáëåíèÿ êèñëîðîäà ó ãèïåðòèðåîèäíîé ãðóïïû æèâîòíûõ (ýëåêòðîííûé ãàçîàíàëèçàòîð «Radiometer») áûëà íà 5,0 ± 0,58 ìë/êã/ ìèí áîëüøå (P < 0,01) óðîâíÿ êîíòðîëÿ, à ðåêòàëüíàÿ òåìïåðàòóðà ïîâûñèëàñü äî 38,5 ± 0,10Ñ, ïðåâûøàÿ êîíòðîëüíûé óðîâåíü íà 0,7 ± 0,10Ñ (P < 0,05). Òàêîé òèðåîèäíûé ñòàòóñ ìîæíî îöåíèòü, êàê ñîñòîÿíèå ëåãêîãî ãèïåðòèðåîçà [1, 8].  õîäå îïûòà ó íàðêîòèçèðîâàííîãî æèâîòíîãî (òèîïåíòàë â äîçå 75 ìã/êã âíóòðèáðþøèííî) ïðåïàðèðîâàëè ìàëîáåðöîâûé íåðâ â îáëàñòè áåäðà (íà ðàññòîÿíèè 1 ñì îò êîëåííîãî ñóñòàâà) è ïîäâîäèëè ïîä íåãî ýëåêòðîäû. Äëÿ îòâåäåíèÿ áèîïîòåíöèàëîâ îò ïåðåäíåáåðöîâîé ìûøöû èñïîëüçîâàëè áèïîëÿðíûå èãîëü÷àòûå ñòàëüíûå ýëåêòðîäû ñ ìåæýëåêòðîäíûì ðàññòîÿíèåì 1 ìì, êîòîðûå ââîäèëè â ñðåäíþþ ÷àñòü ìûøöû (íà ðàññòîÿíèè 1 ñì îò êîëåííîãî ñóñòàâà). Óñèëåíèå âûçâàííûõ áèîïîòåíöèàëîâ îñóùåñòâëÿëè ñ ïîìîùüþ ýëåêòðîìèîãðàôè÷åñêîãî áèîóñèëèòåëÿ, ïîñòðîåííîãî íà îñíîâå èíñòðóìåíòàëüíîãî äèôôåðåíöèàëüíîãî óñèëèòåëÿ, ãèðàòîðà, ñëóæàùåãî äëÿ ðåæåêöèè ñèãíàëà ýëåêòðîñåòåâîé ïîìåõè (÷àñòîòíûé ðåæåêòîðíûé ôèëüòð), è ãàëüâàíè÷åñêîé îïòðîííîé ðàçâÿçêè. Ýëåêòðîñòèìóëÿòîð ïîçâîëÿë â øèðîêèõ ïðåäåëàõ çàäàâàòü òðåáóåìûå ïàðàìåòðû ïðÿìîóãîëüíûõ èìïóëüñîâ: ÷àñòîòó, äëèòåëüíîñòü, àìïëèòóäó è ïðîäîëæèòåëüíîñòü íàíåñåíèÿ ðàçäðàæåíèÿ. Íà ïåðâîì ýòàïå îïûòà ïðîâîäèëàñü çàïèñü ïåðâûõ äåâÿòè Ì-îòâåòîâ. Ñ ó÷åòîì òîãî îáñòîÿòåëüñòâà, ÷òî Ì-îòâåò ïðåäñòàâëÿåò ñîáîé ñóììàðíûé ïîòåíöèàë ìûøöû, îïðåäåëÿåìûé ÷èñëîì è ñèíõðîíèçàöèåé âîçáóäèâøèõñÿ ìûøå÷íûõ âîëîêîí, òî äëÿ ãåíåðàöèè ïîëíîìàñøòàáíîãî Ì-îòâåòà ïðèìåíÿëèñü èìïóëüñû ñòèìóëÿòîðà àìïëèòóäîé 2 ðåîáàçû è äëèòåëüíîñòüþ 2 õðîíàêñèè ïðè ÷àñòîòå 5 èìï/ñ. Òàêèå êîëè÷åñòâåííûå ïàðàìåòðû èìïóëüñà ïðåâûøàþò ïîðîãîâûå, íî îñòàþòñÿ ñóùåñòâåííî ìåíüøå ñóáìàêñèìàëüíûõ ñòèìóëîâ [1]. Íà âòîðîé ñòàäèè ýêñïåðèìåíòà ðåãèñòðèðîâàëàñü îöèôðîâàííàÿ ìèîãðàììà (ðèñ. 1) ïðè èçîòîíè÷åñêîì ñîêðàùåíèè (èìïóëüñû ñòèìóëÿòîðà äëèòåëüíîñòüþ 100 ìêñ ïðè ÷àñòîòå 80 Ãö, ïðîäîëæèòåëüíîñòü ðàçäðàæåíèÿ ñîñòàâèëà 6 ñ), ÷òî ïîçâîëÿëî âû÷èñëèòü ëàòåíòíûé ïåðèîä ìåòðè÷åñêîãî óêîðî÷åíèÿ (ñîêðàùåíèÿ) ìûøöû (ËÏÑ) è ðàññ÷èòàòü â äàëüíåéøåì ñêîðîñòíûå ïàðàìåòðû åå ñîêðàùåíèÿ (ðèñ. 2).  êîíöå îïûòà æèâîòíûå çàáèâàëèñü ïóòåì äåêàïèòàöèè, ïîñëå ÷åãî äëÿ ïîñëåäóþùåãî îïðåäåëåíèÿ êîíöåíòðàöèè öèðêóëèðóþùåãî ñâîáîäíîãî òðèéîäòèðîíèíà ïðîèçâîäèëñÿ çàáîð êðîâè. Îïðåäåëåíèå ãîðìîíà ïðîâîäèëîñü ñ ïîìîùüþ ìåòîäà èììóíîôåðìåíòíîãî àíàëèçà ñ èñïîëüçîâàíèåì ñèñòåìû ôèðìû ‘’ThermoLabsystems’’ è ñòàíäàðòíûì íàáîðîì ðåàãåíòîâ ÒèðîèäÈÔÀ-òðèéîäòèðîíèí ñâîáîäíûé (Ðîññèÿ). Ïîëó÷åííûå äàííûå îáðàáàòûâàëèñü ñ ïîìîùüþ ñòàíäàðòíûõ ìåòîäîâ âàðèàöèîííîé ñòàòèñòèêè. Ðåçóëüòàòû èññëåäîâàíèé è èõ îáñóæäåíèå. Ðåçóëüòàòû ýêñïåðèìåíòîâ ïîêàçàëè, ÷òî â êîíòðîëüíîé ãðóïïå êðûñ óðîâåíü ñâîáîäíîãî òðèéîäòèðîíèíà ñîñòàâëÿë 4,46 ± 0,17 ïìîëü/ë, à ýêçîãåííîå ââåäåíèå òðèéîäòèðîíèíà âûçûâàëî ïîâûøåíèå åãî êîíöåíòðàöèè äî 11,46 ± 0,27 ïìîëü/ë (+157 %, ð < 0,001). Õàðàêòåð ðàñïðåäåëåíèÿ âàðèàöèîííûõ ðÿäîâ äëÿ Ò3ñâîá. ó êðûñ êîíòðîëüíîé è ãèïåðòèðåîèäíîé ãðóïï íà îñíîâàíèè òåñòà Øàïèðî-Óèëêà îïðåäåëÿëñÿ íîðìàëüíûì çàêîíîì (ð = 0,103 è ð = 0,213, ò. å. íóëåâàÿ ãèïîòåçà ñîõðàíÿëàñü). Àíàëèç õàðàêòåðà çàâèñèìîñòè äëèòåëüíîñòè ëàòåíòíîãî ïåðèîäà âîçáóæäåíèÿ (ËÏÂ) ìûøöû îò óðîâíÿ öèðêóëèðóþùåãî òðèéîäòèðîíèíà ïîêàçàë ñëåäóþùåå (ðèñ. 3, òàáë.). Ïðè ýêñïåðèìåíòàëüíîì ãèïåðòèðåîçå èñïîëüçîâàííîé ìîäåëè ËÏÓ, âî-ïåðâûõ, óêîðà÷èâàåòñÿ (íà 12 %), à âî-âòîðûõ, âñå åùå îñòàåòñÿ ÷óâñòâèòåëüíûì ê äîïîëíèòåëüíîé ìîäóëÿöèè ïàòîôèçèîëîãè÷åñêèì êîíöåíòðàöèÿìè òðèéîäòèðîíèíà. Òðåòüèì ïîêàçàòåëåì, îòðàæàþùèì ôóíêöèîíàëüíîå ñîñòîÿíèå ñêåëåòíîé ìûøöû, ÿâëÿåòñÿ ïàðàìåòð «ñêîðîñòü ðàçâèòèÿ ñîêðàùåíèÿ» (ðèñ. 5). Ðåçóëüòàòû ðàñ÷åòà ñêîðîñòè óêîðî÷åíèÿ ïåðåäíåáåðöîâîé ìûøöû ïðè èçîòîíè÷åñêîì ñîêðàùåíèè ñ ãðóçîì 20 ã ïîêàçàëè ñëåäóþùåå. Âî-ïåðâûõ, ñêîðîñòü ðàçâèòèÿ ñîêðàùåíèÿ ìûøöû ó æèâîòíûõ îáåèõ ãðóïï â ïðîöåññå ñîêðàòèòåëüíîãî öèêëà ñóùåñòâåííî ðàçëè÷àëàñü. Íà îñíîâàíèè ðàñ÷åòîâ ìîæíî âûäåëèòü, ïî êðàéíåé ìåðå, òðè ó÷àñòêà íà øêàëå âðåìåíè. Ïåðâàÿ ôàçà õàðàêòåðèçîâàëàñü íåáîëüøîé, íî áûñòðî íàðàñòàþùåé ñêîðîñòüþ ñîêðàùåíèÿ, è ïðîäîëæàëàñü ó êðûñ îáåèõ ãðóïï äî çíà÷åíèÿ â îáëàñòè 30 ìñ. Íàèáîëüøàÿ ñêîðîñòü îòìå÷àëàñü âî âòîðîé, áûñòðîé, ôàçå ñîêðàòèòåëüíîãî àêòà.  äàëüíåéøåì ñêîðîñòü ñîêðàùåíèÿ ìûøöû ó êðûñ îáåèõ ãðóïï ðåçêî ïàäàëà ïî çàêîíó ñòåïåííîé ôóíêöèè: ó = 34481õ-2,84 (êîíòðîëü) è ó = 2034õ-2,149 (ãèïåðòèðåîç). Âî-âòîðûõ, õîòÿ îáùèé õàðàêòåð äèíàìèêè ñêîðîñòè ñîêðàùåíèÿ ìûøöû ó êðûñ îáåèõ ãðóïï íå ðàçëè÷àëñÿ, îäíàêî àáñîëþòíàÿ âåëè÷èíà ïàðàìåòðà «ñêîðîñòü ñîêðàùåíèÿ» ó êðûñ ãèïåðòèðåîèäíîé ãðóïïû áûëà çíà÷èòåëüíî âûøå, äîñòèãàÿ óðîâíÿ 0,12 ± 0,018 ì/ñ (ó êîíòðîëÿ 0,0894 ± 0,0087 ì/ñ, ò. å. íà 34 % íèæå, p < 0,05). Â-òðåòüèõ, ïðè ýêñïåðèìåíòàëüíîì ãèïåðòèðåîçå íàáëþäàëîñü ñìåùåíèå ïèêà ñêîðîñòè ñîêðàùåíèÿ ìûøöû ïî øêàëå âðåìåíè â ìåíüøóþ ñòîðîíó (102 %, p < 0,01). Òàê, åñëè ó êðûñ êîíòðîëüíîé ãðóïïû ìàêñèìàëüíàÿ ñêîðîñòü îòìå÷åíà íà 49,8 ± 2,49 ìñ, òî ó æèâîòíûõ ãèïåðòèðåîèäíîé ãðóïïû ñîîòâåòñòâåííî íà 100,6 ± 3,8 ìñ. Äåëàåòñÿ îáùåå çàêëþ÷åíèå î òîì, ÷òî ñâîáîäíûé öèðêóëèðóþùèé òðèéîäòèðîíèí â øèðîêîì äèàïàçîíå êîíöåíòðàöèè «2,0 – 13,8 ïìîëü/ë» ÿâëÿåòñÿ ìîùíûì ìîäóëÿòîðîì ïðîöåññà âîçáóæäåíèÿ ñêåëåòíîé ìûøöû è îñíîâíûõ ïàðàìåòðîâ ñêîðîñòè åå ñîêðàùåíèÿ. Îáúÿñíåíèå ïîëó÷åííûõ ðåçóëüòàòîâ ìîæåò áûòü ñäåëàíî ñ ó÷åòîì èìåþùèõñÿ â ëèòåðàòóðå äàííûõ îòíîñèòåëüíî íàïðàâëåííîñòè äåéñòâèÿ òèðåîèäíûõ ãîðìîíîâ íà ìîðôîëîãè÷åñêèå, ýëåêòðîôèçèîëîãè÷åñêèå è áèîõèìè÷åñêèå ïîêàçàòåëè ñêåëåòíîé ìûøöû. Òàê, â ëèòåðàòóðå ñóùåñòâóþò ñîîáùåíèÿ â ïîëüçó ïîçèòèâíîãî âëèÿíèÿ ãèïåðòèðåîèäíûõ ñîñòîÿíèé íà ïëîòíîñòü Nà+-êàíàëîâ â ïëàçìàòè÷åñêîé ìåìáðàíå [13], à òàêæå íà àêòèâíîñòü è êîíöåíòðàöèþ ìîëåêóë Ñà2+-ÀÒÔàçû â ìåìáðàíàõ ñàðêîïëàçìàòè÷åñêîãî ðåòèêóëþìà [10, 11, 14]. Òàêèì îáðàçîì, åñëè äîïóñòèòü âåðîÿòíîñòü îñóùåñòâëåíèÿ êàêèõ-ëèáî èç óïîìÿíóòûõ âûøå ñòèìóëèðóþùèõ ýôôåêòîâ òèðåîèäíûõ ãîðìîíîâ íà ñîñòîÿíèå Ñà2+-íàñîñà ñàðêîïëàçìàòè÷åñêîãî ðåòèêóëþìà, ñðîäñòâî ìèîôèáðèëëÿðíûõ áåëêîâ ê èîíàì Ñà2+, àêòèâíîñòü ìèîçèíîâîé ÀÒÔàçû, ïàññèâíûé âûõîä Ñà2+ â ñàðêîïëàçìó âîëîêíà, òî çàêîíîìåðíî ìîæíî îæèäàòü èçìåíåíèÿ ñêîðîñòíûõ õàðàêòåðèñòèê ñêåëåòíîé ìóñêóëàòóðû ïîä âëèÿíèåì ôèçèîëîãè÷åñêèõ è ïîâûøåííûõ êîíöåíòðàöèé éîäòèðîíèíîâ.  ñâÿçè ñ îòìå÷åííûì, íàèáîëåå âåðîÿòíûì îáúÿñíåíèåì óõóäøåíèÿ ñêîðîñòíûõ õàðàêòåðèñòèê èññëåäóåìîé ìûøöû æèâîòíûõ ïðè ñâåðõôèçèîëîãè÷åñêèõ óðîâíÿõ öèðêóëèðóþùåãî òðèéîäòèðîíèíà, íà íàø âçãëÿä, ÿâëÿåòñÿ äîïóùåíèå î òîì, ÷òî áîëüøàÿ, ïî ñðàâíåíèþ ñ êîíòðîëåì, ÷àñòü ìûøå÷íûõ âîëîêîí ïðîñòî âûêëþ÷àåòñÿ èç ñîêðàòèòåëüíîãî àêòà ïî ïðè÷èíå êàêèõ-òî ôóíêöèîíàëüíûõ, ìåòàáîëè÷åñêèõ èëè äðóãèõ èçìåíåíèé. Âûâîäû. 1.  äèàïàçîíå ôèçèîëîãè÷åñêèõ êîëåáàíèé óðîâíÿ ñâîáîäíîãî òðèéîäòèðîíèíà (2,0 – 7,0 ïìîëü/ë) ñóùåñòâóåò îòðèöàòåëüíàÿ çàâèñèìîñòü ìåæäó åãî êîíöåíòðàöèåé è äëèòåëüíîñòüþ ëàòåíòíîãî ïåðèîäà ãåíåðàöèè Ì-îòâåòà ñêåëåòíîé ìûøöû ïðè âûñîêîì çíà÷åíèè êîýôôèöèåíòà êîððåëÿöèè Ïèðñîíà (-0,88); ó æèâîòíûõ ãèïåðòèðåîèäíîé ãðóïïû çàâèñèìîñòü ìåæäó óðîâíåì öèðêóëèðóþùåãî ñâîáîäíîãî òðèéîäòèðîíèíà (â äèàïàçîíå êîíöåíòðàöèè îò 8,8 äî 13,8 ïìîëü/ë) è äëèòåëüíîñòüþ ëàòåíòíîãî ïåðèîäà âîçáóæäåíèÿ ìûøöû îòñóòñòâîâàëà. 2. Ñðåäíÿÿ âåëè÷èíà äëèòåëüíîñòè ëàòåíòíîãî ïåðèîäà ìåòðè÷åñêîãî ñîêðàùåíèÿ ïåðåäíåáåðöîâîé ìûøöû ó êðûñ ãèïåðòèðåîèäíîé ãðóïïû áûëà íà 12 % ìåíüøå óðîâíÿ êîíòðîëÿ. 3. Ìåæäó óðîâíåì ñâîáîäíîãî òðèéîäòèðîíèíà è çíà÷åíèåì ëàòåíòíîãî ïåðèîäà ñîêðàùåíèÿ ìûøöû ñóùåñòâóåò îòðèöàòåëüíàÿ ñâÿçü; îäíàêî â äèàïàçîíå ñâåðõôèçèîëîãè÷åñêîé êîíöåíòðàöèè òðèéîäòèðîíèíà «8,8 – 13,8 ïìîëü/ë» òàêàÿ ñâÿçü îñëàáëÿåòñÿ (67 %). 4. Àáñîëþòíàÿ âåëè÷èíà ñêîðîñòè ñîêðàùåíèÿ ìûøöû (ì/ñ) ó êðûñ ãèïåðòèðåîèäíîé ãðóïïû áûëà íà 34 % íèæå óðîâíÿ êîíòðîëÿ ïðè îäíîâðåìåííîì ñìåùåíèè åå ïèêà ïî øêàëå âðåìåíè â ìåíüøóþ ñòîðîíó (102 %). Âèÿâëåíî, ùî ì³æ ð³âíåì â³ëüíîãî òðèéîäòèðîí³íó ³ çíà÷åííÿì ëàòåíòíîãî ïåð³îäó ñêîðî÷åííÿ ì’ÿçà ³ñíóº íåãàòèâíèé çâ’ÿçîê; ïðîòå â ä³àïàçîí³ íàä ô³ç³îëîã³÷íî¿ êîíöåíòðàö³¿ òðèéîäòèðîí³íó «8,8 – 13,8 ïìîëü/ë» òàêèé çâ’ÿçîê ïîñëàáëþºòüñÿ (67 %). Àáñîëþòíà âåëè÷èíà øâèäêîñò³ ñêîðî÷åííÿ ì’ÿçà ó ùóð³â ã³ïåðòèðåî¿äíî¿ ãðóïè áóëà íà 34 % íèæ÷å ð³âíÿ êîíòðîëþ ïðè îäíî÷àñíîìó çì³ùåíí³ ¿¿ ï³êó çà øêàëîþ ÷àñó â ìåíøó ñòîðîíó (102 %). Ðîáèòüñÿ çàãàëüíèé âèñíîâîê ïðî òå, ùî â³ëüíèé öèðêóëþþ÷èé òðèéîäòèðîí³í â ä³àïàçîí³ ô³ç³îëîã³÷íî¿ êîíöåíòðàö³¿ ³ çà ¿¿ ìåæàìè «äî 13,8 ïìîëü/ë» º ïîòóæíèì ìîäóëÿòîðîì ïðîöåñó çáóäæåííÿ ñêåëåòíîãî ì’ÿçà òà îñíîâíèõ ïàðàìåòð³â øâèäêîñò³ éîãî ñêîðî÷åííÿ. Âûÿâëåíî, ÷òî ìåæäó óðîâíåì ñâîáîäíîãî òðèéîäòèðîíèíà è çíà÷åíèåì ëàòåíòíîãî ïåðèîäà ñîêðàùåíèÿ ìûøöû ñóùåñòâóåò îòðèöàòåëüíàÿ ñâÿçü; îäíàêî â äèàïàçîíå ñâåðõôèçèîëîãè÷åñêîé êîíöåíòðàöèè òðèéîäòèðîíèíà «8,8 – 13,8 ïìîëü/ë» òàêàÿ ñâÿçü îñëàáëÿåòñÿ (67 %). Àáñîëþòíàÿ âåëè÷èíà ñêîðîñòè ñîêðàùåíèÿ ìûøöû ó êðûñ ãèïåðòèðåîèäíîé ãðóïïû áûëà íà 34 % íèæå óðîâíÿ êîíòðîëÿ ïðè îäíîâðåìåííîì ñìåùåíèè åå ïèêà ïî øêàëå âðåìåíè â ìåíüøóþ ñòîðîíó (102 %). I. Abstract. Despite a fairly complete characterization of the effect of thyroid hormones on different sides of the functional state of skeletal muscle, a lot of aspects remain poorly studied. First of all it concerns the nature of the dependence between the degree of distortion of thyroid status and the intensity of pathophysiological mechanisms, in developing neuromuscular system. The aim of this work was in the conditions in situ to elucidate the nature of the relations between indicators of contraction mobile parameters of the anterior large tibial muscle of white rats of different thyroid status, on the one hand, and the level of circulating free triiodothyronine, on the other hand. The experiments were performed on 80 adult white rats-males. The first group of rats (30 animals) were injected subcutaneously by triiodothyronine in the dose of 15 mg/kg every day during 4 days (hyperthyroid group). The second group of rats (50) was the control. Under the conditions in situ during the experiments the latent period of the M-response generation of anterior tibial muscle, the latent period of its metrical shortening were measured, the speed of muscle contraction was calculated. At the end of the experiment the animals were killed by decapitation, then blood sampling was collected for the subsequent determination of the concentration of circulating free triiodothyronine. The hormone determination was carried out by means of the method of enzyme-linked immunoassay with the use of the system of the firm “ThermoLabsystems” and a standard set of reagents Thyroid ELISA free triiodothyronine (Russia). It is established that within the range of physiological fluctuations in the level of free triiodothyronine (2.0 – 7.0 pmol/l) there is a negative relationship between the concentration and the duration of the latent period of the M-response generation of skeletal muscle at high value of Pearson correlation coefficient (-0,88); hyperthyroid group animals’ relationship between the level of circulating free triiodothyronine (in the concentrations ranging from 8.8 to 13.8 pmol/l) and the duration of latent period of excitation of the muscle was absent. It is shown that the average duration of the latent period of the metric contraction of hyperthyroid group rats’ anterior tibial muscle was 12 % less than the control level. It is revealed that there is a negative relationship between the level of free triiodothyronine and the term of the latent period of muscle contraction; however, in the range superphisiological concentration of triiodothyronine “8,8 – of 13.8 pmol/l” this relationship is weakened (67 %). The absolute value of the speed of muscle contraction (m/s) of hyperthyroid group rats was 34 % below the control level with a simultaneous shift of its peak on the time scale in the lower side (102 %). The general conclusion that free circulating triiodothyronine in the range of physiological concentration and beyond it “to 13.8 pmol/l” is a powerful modulator of the process of excitation of skeletal muscle and of the main parameters of its contraction speed.
